I am a huge advocate of Ableton Live. Its not cheap, but if you're serious about computer DJ'ing, then it is well worth it. Personally, I use it for programming, not DJ'ing, but I know that it has a good following of DJs too. Go to http://www.ableton.com and check it out for yourself.

There is no doubt in my mind that Ableton is the best affordable software out there. Sure, the professionals use ProTools and other bigtime software, but many artists across the world use Ableton Live for live performances, productions and compositions, and much more. Fruity Loops is more of a toy for beginners rather than something powerful that you can make good songs and remixes and mixes in.

    Ableton Live! 6 for producing
Traktor 3 for mixing (built in final-scratch software)

Secondarily, Cubase is good, but requires hardware, and I've heard good things about Reaktor, but haven't tried it. Fruityloops is pretty out-dated.
